Inputting Or Choosing Sequences
Select one of the following methods to enter your desired sequence:
“Paste Sequence”: cut and paste your sequence into the box. Either plain DNA sequence or a FASTA
formatted sequence is acceptable. Alternatively, a short sequence can be typed in directly.
“Upload File”: Click “Browse” and select the desired file from your computer.
“Use GenBank”: Enter a valid Genbank number (If you do not know the GenBank number of your sequence,
Click the GenBank link).
“Select Plasmid Vector”: Select from a set of pre-populated plasmid vectors by using the pulldown menu.
Alternatively, start typing in the name of your plasmid and it will come up if available.
“Select Viral & Phage”: Select from a set of pre-populated viral and phage sequences by using the
pulldown menu. Alternatively, start typing in the name and it will come up if available.
Setting Preferences
Check “Circular” if your sequence is circular. If not, please leave blank.
Click “More” to see further options, including: to add other enzyme Types (I and/or III, homing and
nicking), to specify one or more methylation sensitivities, to specify minimum ORF length, to change the
genetic code to be used, or to specify a sub-region within the input file. You can also define up to 40
oligonucleotide sequences of your choice.
Naming Projects
If you wish to identify the input sequence by a specific name, enter it here. If left blank, your sequence
will be named “Default” and will be overwritten with each new project.
